Prevents joint destruction by reducing osteoclast activity.

Leflunomide is an immunomodulatory agent with anti-inflammatory and antiproliferative properties. It is a prodrug, rapidly converted in the body to its active metabolite teriflunomide (A77 1726), which inhibits the mitochondrial enzyme dihydroorotate dehydrogenase (DHODH). This enzyme is essential for pyrimidine synthesis, which is crucial for activated T-cell proliferation.
